| Summary | Exemestane is an orally active irreversible steroidal aromatiase inhibitor used for the therapy of metastatic postmenopausal breast cancer.The objective of this work was to develop and validate a highly sensitive on-line extraction LC-MS/MS method for the determination of exemestane (EXE) and 17β-hydroxy- exemestane (17β EXE) in human plasma.. A high sensitive, accurate and robust method for the determination of exemestane and its metabolite 17β-OH exemestane was developed. The study also demonstrates how the use of selective analyte isolation techniques, chromatography and MS/MS detection make it possible to analyse steroidal compounds susceptible to isobaric interferences at trace levels. |
